Prof (Brain, nella versione originale) è un topo di laboratorio geneticamente modificato: egli è dotato di un'incredibile intelligenza ed il suo unico scopo è la conquista del mondo. Per far questo, si avvale dell'aiuto del suo assistente Mignolo (Pinky, nella versione originale), un altro topo geneticamente modificato; quest'ultimo è assai tontolone, pasticcione e confusionario, ma, nonostante tutto, dopo ogni tentativo fallito, aiuta Prof a vedere la vita un po' meno grigia.

Spin-off degli Animaniacs, le vicende di questo cartone - prodotto anch'esso da Steven Spielberg - ha avuto un seguito in cui Mignolo e Prof vengono affiancati da un nuovo personaggio: Pinky, Elmyra and the Brain.
